The WXX(F/Y) peptide repeats of Pex5p are important determinants for Pex14p binding

Exponentially growing cells (yeast strain SFY526), transformed with plasmids encoding one of the bait proteins (PTS1, Pex12p(275–359), or Pex14p, fused to the Gal4p DNA-binding domain) and one of the prey proteins (Pex5p(222–602), Pex5p(222–602)(W271G), or Pex5p(332–602), fused to the Gal4p activation domain), were assayed for β-galactosidase using o-nitrophenyl-β-d-galactopyranoside as substrate (see “Experimental Procedures”). The optical densities were measured at 420 nm, corrected for the blank (“empty” plasmids), and normalized for culture densities (optical density at 600 nm = 10) and time (24 h). The values given are the mean (± the S.D.) of at least three measurements performed on cultures derived from independent single yeast colonies.
